摘要
目的:观察人工肾小管辅助装置(bio-artificialrenaltubuleassistdevice,RAD)对动物模型细胞因子水平、生存时间的影响。方法:经盲肠结扎加穿孔及双侧输尿管结扎术造模后的12头猪被随机平均分为3组,分别为RAD治疗组、不含小管细胞的假RAD治疗组和非治疗对照组。各组分别定时抽取静脉血检测血液电解质、肾功能指标。应用猪特异性酶联免疫试剂盒测定肿瘤坏死因子-α(TNF-α)水平,并记录两组动物的存活时间。结果:RAD组透析前后的血钾均值为(6.92±0.12),(4.39±0.46)mmol/L,血肌酐均值为(911.3±100.7),(652±130)μmol/L,TNF-α均值为(0.537±0.038),(0.278±0.069)μg/L,与对照组比较,差异均有显著性意义(P<0.05)。RAD组的平均生存时间为犤(110.25±18.69)h犦,明显长于非治疗对照组犤(72.84±11.62)h犦和假RAD治疗组犤(75.20±19.08)h犦,差异有显著性意义(P<0.05)。结论:用RAD治疗可降低多脏器功能障碍综合征(multipleorgandys-functionfailure,MODS)并急性肾功能衰竭(acuterenalfailure,ARF)动物的TNF-α水平,并延长其存活时间。
AIM:To observe the effect of bio-artificial renal tubule assist device (RAD) on the cytokine and survival time in porcine models. METHODS:Twelve porcine models were divided into three groups after underwent c ecal ligation and perforation (CLP) plus bilateral ureteral ligation:RAD group, sham RAD group (without tubule cells in the filter) and control group (no treatm ent). Venous blood was drawn at different time points to determine blood electro lure and renal function parameters. The tumor necrosis factor-α(TNF-α) conce ntration was measured by using specific enzyme-linked immunoadsordent kit, mean while, the survival time was also observed and compared between these groups. RESULTS: The value of blood concentrations of kalium [(6.92±0.12), (4.39±0.4 6) mmol/L],creatinine [(911.3±100.7),(652±130) μmol/L] and TNF-α[(0.537±0. 038), (0.278±0.069) μg/L] before and after dialysis in the RAD group were sign ificantly different from those in the control group (P< 0.05). The average survi val time in the RAD group was (110.25±18.69) hours, distinctly longer than thos e in the control group [(72.84±11.62) hours] and sham RAD group [(75.20±19.08) hours] (P< 0.05). CONCLUSION:RAD treatment can reduce the blood level of TNF-αof model with mu ltiple organ dysfunction failure (MODS) complicated by acute renal failure (ARF) ,and the survival time can also be extended.
