摘要
目的 研究中药薏苡仁酯对喉癌Hep 2细胞的增殖抑制作用及机制。方法 应用MTT法测定不同浓度的薏苡仁酯对Hep 2细胞增殖抑制情况 ;应用单细胞凝胶电泳法 ,检测薏苡仁酯引起Hep 2细胞的DNA损伤情况。结果 5 μl/ml～ 4 0 μl/ml的薏苡仁酯作用 4 8h后明显抑制Hep 2细胞的增殖作用 (P <0 .0 1) ;5 μl/ml和 10 μl/ml的薏苡仁酯作用 4 8h后引起Hep 2细胞DNA损伤 (P <0 .0 1)。结论 在体外培养的条件下中药薏苡仁酯能明显抑制Hep 2细胞的增殖 。
Objective To study the inhibitory effects and mechanism of the traditional Chinese drug Coixlipid on laryngeal carcinoma cells Hep 2.Methods The inhibitory effect of Coixlipid on the growth of Hep 2 cell was evaluated by MTT assay.The single cell gel electrophoresis(SCGE) was used to detect the DNA damage of Hep 2 cells by Coixlipid.Results After Hep 2 cell had been treated with Coixlipid (5μl/ml～40μl/ml) for 48h,the growth of Hep 2 cell was significantly inhibited(P<0.01).After the cell had been treated with Coixlipid (5μl/ml and 10μl/ml) for 48h,its DNA was damaged(P<0.01).Conclusion The traditional Chinese drug Coixlipid can effectively inhibit the growth of Hep 2 cell in vitro,its mechanism is related to the DNA damage.
