摘要
目的 建立人血清脂蛋白 (a) [Lp(a) ]循环免疫复合物 (CIC)酶联免疫吸附试验 (ELISA)检测法 ,探讨人体内是否存在Lp(a) CIC及其临床价值。 方法 分别建立抗人IgG为包被抗体 ,酶标抗载脂蛋白 (a)和抗载脂蛋白B为检测抗体的Lp(a) CIC、低密度脂蛋白 (LDL) CICELISA检测法 ,并对16 0例冠心病患者和 2 90名对照人群进行分析。结果 两组相比 ,冠心病患者Lp(a) CIC ( 2 2 4±1 71AUvs 1 6 2± 1 5 0AU)、LDL CIC ( 2 77± 1 2 9AUvs 1 4 0± 0 92AU)水平均显著升高 (P <0 0 0 01)。LDL CIC水平分别与血浆LDL、胆固醇 (r =0 5 4 9)、载脂蛋白B (r =0 4 78)、总胆固醇 (r =0 4 13)、甘油三酯 (r=0 2 4 8)和Lp(a) (r=0 2 39)浓度正相关 (均P <0 0 0 0 1) ,与高密度脂蛋白、胆固醇 (r =- 0 30 8)和载脂蛋白A1(r=- 0 2 0 9)浓度负相关 (P <0 0 0 0 1)。Lp(a) CIC水平分别与血浆Lp(a) (r =0 5 77,P <0 0 0 0 1)、LDL胆固醇 (r =0 2 0 7,P <0 0 0 0 1)、载脂蛋白B (r =0 12 8,P <0 0 1)和总胆固醇 (r=0 14 5 ,P <0 0 1)浓度正相关 ,与高密度脂蛋白、胆固醇 (r=- 0 12 6 ,P <0 0 1)和载脂蛋白A1(r=- 0 0 96 ,P <0 0 5 )浓度负相关。LDL CIC与Lp(a) CIC水平亦相关 (r =0 313,P
Objective To make sure the existence of IgG bound Lp(a) circulating immune complexes (CIC) [Lp(a) CIC] and to investigate its significance as a risk factor for the development of atherosclerosis Methods Two “sandwich” ELISAs for measuring plasma Lp(a) CIC and LDL CIC level were developed by use of anti human IgG(Fab) as the capture antibody and anti apo(a) or anti apoB monoclonal antibody as the enzyme conjugate The Lp(a) CIC and LDL CIC levels were studied in both 160 patients with coronary heart disease (CHD) and 290 control subjects Results Both plasma Lp(a) CIC (2 24±1 71AU vs 1 62±1 50AU) and LDL CIC (2 77±1 29AU vs 1 40±0 92AU) levels in patients with CHD were significantly higher than those of control ( P <0 000 1) The relationships between Lp(a) CIC, LDL CIC levels and other lipid traits in all the studied subjects ( n =450) were carried out LDL CIC levels as positively correlated with LDL C ( r =0 549, P <0 000 1), apoB ( r =0 478, P <0 000 1), TC ( r =0 413, P <0 000 1), TG ( r =0 248, P <0 000 1), and Lp(a) ( r =0 239, P <0 000 1) concentrations, while negatively correlated with HDL C ( r =-0 308, P <0 000 1) and apoAI ( r = 0 209, P <0 000 1) concentrations, respectively Similarly, Lp(a) CIC levels were positively correlated with Lp(a) ( r =0 577, P <0 000 1), LDL C ( r =0 207, P <0 000 1), apoB ( r =0 128, P <0 01), and TC ( r =0 145, P <0 01) concentrations, while negatively correlated with HDL C ( r =-0 126, P <0 01) and apoAI ( r =-0 096, P <0 05) concentrations, respectively Furthermore, a significantly positive relation between LDL CIC and Lp(a) CIC levels was also found ( r =0 313, P <0 000 1) Conclusion The study confirms the existence of Lp(a) IC in both the plasma of patients with CHD and control subjects And Lp(a) CIC level increases in the CHD patients
