摘要
目的 :探讨宽叶缬草对局灶性脑缺血的影响。方法 :6 0只健康、雄性Wistar大鼠分为正常组、假手术组、大脑中动脉闭塞组 (MCAO组 )、对照组 (NS组 )、宽叶缬草组 (VOL组 ) ,制作可复流大脑中动脉闭塞大鼠模型。NS组、VOL组大鼠大脑中动脉闭塞后 2h分别以生理盐水及宽叶缬草灌胃 ,每 8h 1次 ;术后第 5d进行Nissl、TTC染色及脑梗死体积比测定。结果 :VOL组的脑梗死体积比显著低于MCAO组 ;VOL组海马各区及大脑皮层神经元损伤轻 ,丢失相对较少。结论 :宽叶缬草可以减轻局灶性脑缺血后脑组织病理学损害 ,具有脑保护作用。
Objective: To evaluate the effect of Valeriana officinalis var latifolia miq (VOL) on brain injury in experimental reversible middle cerebral artery occlusion (MCAO). Methods: Eighty male Wistar rats were randomly divided into 5 groups: normal group, sham operation group, MCAO group, MCAO+normal saline group (NS group) and MCAO+VOL group (VOL group). MCAO rats were induced by intraluminal suture occlusion. The drugs, such as VOL or normal saline, were administered through gastrogavage at second h after MCAO in NS group and VOL group, once every 8 h. The rats were sacrificed on 5th day after MCAO. Then Nissl staining and TTC staining were performed. The numerical density (ND) of Nissl staining neurons and the percent of infarction volume in hemisphere volume were measured by CIAMS image analysis system. Results: The percent of infarction volume in hemisphere volume was markedly reduced in VOL group (31.34±6.55) as compared with MCAO group (48.97± 7.21). The ND of Nissl staining neurons in hippocampal and cortex were decreased significantly after MCAO, and the neuron loss was greatly alleviated after the treatment with VOL. Conclusion: VOL could attenuate the neuron damage induced by MCAO and play a brain protective role against ischemic injury.
