摘要
目的:用荧光光谱技术研究在生理条件下氧氟沙星与DNA分子间结合作用的机理。方法:在pH 7.20条件下,测量不同DNA浓度下氧氟沙星的荧光激发与发射光谱。结果:DNA存在能使氧氟沙星荧光猝灭,测得该反应的结合常数K=2448 L·mol-1。结论:氧氟沙星与DNA结合生成了不发光的复合物导致氧氟沙星的荧光猝灭。
Objective:To study the interaction between ofloxacin and DNA in physiological condition by fluorescence spectroscopy. Methods:Fluorescence excitation and emission spectra of 2. 76×10-6mol·L-1 ofloxacin with various concentration of calf thymus DNA at pH 7. 20 buffer solution was measured. Results: Ofloxacin fluorescence intensity was obvious decreased in DNA solution and the binding constant K was found to be 2448 L·mol-1. Conclusion : The interaction between ofloxacin and DNA is strong and the complex formation is due to the ofloxacin fluorescence quenching.
