银杏叶提取物EGb761对实验性变态反应性脑脊髓炎的影响 被引量:7

摘要 目的:探讨银杏叶提取物EGb761对实验性变态反应性脑脊髓炎的免疫机制的影响。方法:以豚鼠全脊髓均浆与福氏完全佐剂为抗原免疫豚鼠诱导实验性变态反应性脑脊髓炎(experimentalallergicencephalomyelitis,EAE)模型。同时以醋酸泼尼松和EGb761分别于免疫当日开始灌胃,连续5周。分为正常对照组、模型组、醋酸泼尼松治疗组和EGb761治疗组,观察EAE症状并评分。全部动物内眦采血,留取血清,采用双抗夹心酶联免疫吸附法(ELISA法)和放免法测白细胞介素12(IL-12),白细胞介素10(IL-10)含量。第5周后全部动物乙醚麻醉后内灌注固定,取出脑、脊髓,以多聚甲醛固定,石蜡包埋、切片,应用免疫组化法测定转化生长因子-β(transforminggrowthfactor-β,TGF-β)、一氧化氮合酶(nitricoxidesynthase,NOS)表达。结果:与模型组相比,醋酸泼尼松治疗组和EGb761治疗组推迟了EAE发病,其临床评分在疾病的起始期、高峰期、缓解后的复发期都明显下降(P<0.05);IL-12含量在模型组、醋酸泼尼松治疗组、EGb761治疗组均较正常对照组明显升高(P<0.05),并且与疾病程度正相关(P<0.05);醋酸泼尼松治疗组和EGb761治疗组与模型组比较IL-10明显上升(P<0.05),TGF-β显著升高(P<0.05),NOS显著下降(P<0.05),且IL-10与TGF-β明显正相关(P<0.05)。 AIM:To explore the effect of ginkgo biloba extract EGb761 on the immunological mechanism of experimental allergic encephalomyelitis(EAE). METHODS:Fresh Guinea pigs spinal cord in saline with an equal volume of complete freuna's adjuvant(CFA) were injected to hind footpads of Guinea pigs as antigen to induce the model of EAE.Meanwhile,prednisone Egb761 were filled into stomach at 0-5 weeks after immunization successively. Animals were divided into normal control group, model group, prednisone group,and EGb761 group to observe EAE symptoms and make score. All animals were taken blood in inner canthus and blood serum was preserved. The concentration of interleukin 12(IL 12) and interleukin 10(IL 10) were measured with enzyme linked immunosorbent assay(ELISA) and radioimmunoassay(RIA).After 5 weeks,all animals were internally fixed after anesthetized with ether,and the brain and spinal cord were taken, and fixed with formaldehydum,and embedded with paraffin, and then slices were made.Expressions of transforming growth factor β(TGF β) and nitric oxide synthase(NOS) were measured with immunohistochemical method. RESULTS:Compared with the model group,the onset of EAE of prednisone group and EGb761 group was delayed,and the clinical scores in the onset phase,peak phase and recurrence phase after remission were significantly lower(P< 0.05).The contents of IL 12 of model group,prednisone group and EGb761 group were significantly higher than that of normal control group(P< 0.05),which were positively correlative with the degree of illness. The contents of IL 10 and the level of TGF of prednisone group and EGb761 group were significantly higher than those of model group(P< 0.05),while the level of NOS was significantly lower(P< 0.05),and IL 10 and TCG βwere significantly in positive correlation(P< 0.05). CONCLUSION:EGb761 has ptotective effects on EAE,and its functional mechanism may have a certain influence on the immunological mechanism of central nervous system.
