蒲金口服液对缺氧缺血性脑损伤新生大鼠一氧化氮合酶的影响 被引量:12

Effects of pujin koufuye on nitric oxide synthase in neonatal rats with hypoxia-ischemia brain damage

摘要 目的:通过蒲金口服液对缺氧缺血性新生大鼠脑组织一氧化氮合酶(nitricoxidosynthase,NOS)的影响,探讨缺氧缺血性脑病的发病机制和化痰活瘀治疗的作用机制。方法:采用7日龄SD大鼠结扎右侧颈总动脉和置入低氧环境中诱发的缺氧缺血性脑损伤模型,于造模后即刻、2,4,6,8和10h,将1日药量共分为6次灌胃犤1.25mg/(kg·d)犦;假手术组术后给予生理盐水灌胃;缺氧缺血性脑损伤模型术后给予生理盐水灌胃;银杏叶提取物组术后给予银杏叶提取物混悬药液灌胃犤40mg/(kg·d)犦,造模后24h处死大鼠,取右侧顶枕区脑组织制成10%脑组织匀浆,用可见光分光光度计通过比色法测定NOS含量。结果:蒲金口服液高剂量组NOS含量为(37.60±5.47)μkat/L,与缺氧缺血性脑损伤模型组NOS含量为(49.97±5.53)μkat/L相比较,差异有极显著性意义(P<0.01);蒲金口服液高剂量组与银杏叶提取物组NOS含量为(42.92±2.70)μkat/L相比较,差异有显著性意义(F=10.22,P<0.05)。结论:蒲金口服液高剂量组能显著降低缺氧缺血性脑损伤大鼠NOS含量,且效果优于银杏叶提取物组。 AIM:To explore the pathogenesis of hypoxic ischemic encephalopathy and functional mechanism of resolving phlem and eliminating statis by detecting the influence of pujin koufuye on nitric oxido synthase(NOS) in the cerebral tissue of rats with hypoxic ischemic encephalopathy.METHODS:Hypoxia ischemia rat models were made in 7 day old SD rats by using the occlusion of right common carotid artery and hypoxia, then pujin koufuye was injected into stomach at 0,2,4,6,8 and 10 hours.The dosages were 2.5 g/kg per day in the high dosage group and 1.25g/kg per day in the low dosage group respectively.The sham operation group and the model group were administrated with normal saline after operation. Tanakan group was filled with tanakan mixed liquid (40 mg/kg per day). After 24 hours,the rats were killed to take out brain tissue in right tip pillow part so as to make 100 g/L homogenate equally plasma,and then the NOS content was mensurated in colorimetry by visible light dispart light photometer.RESULTS:The levels of NOS in high dosage group were(37.60±5.47)μkat/L,which had extremely significant difference from that of model group[(49.97±5.53)μkat/L] (F=0.14,P< 0.01);The levels of NOS in high dosage group was different from that of tanakan group[(42.92±2.70)μkat/L] (F=10.22,P< 0.05). CONCLUSION:Pujin koufuye in the high dosage group can decrease obviously the level of NOS in the cerebral tissue of rats with hypoxic ischemic encephalopathy,the effect of which is better than that of tanakan group.
