染色体标本制备在骨髓MSCs核型鉴定中的研究 被引量:4

摘要 选取对数生长期人骨髓间充质干细胞,用秋水仙素阻断细胞分裂中期,控制秋水仙素的终浓度和作用时间、低渗时间及胰酶消化显带时间,比较染色体标本制备效果。结果显示,秋水仙素的终浓度0.2μg/ml和作用时间6h,低渗时间35min及胰酶消化时间20s时可制备适合的染色体标本,经核型鉴定为正常二倍体细胞。从而探索出骨髓间充质干细胞染色体制备的适合条件,为骨髓间充质干细胞的进一步研究提供了理论依据和实验基础。 After selecting human bone marrow-derived MSCs at the term of logarithmic growth, stopping them at metaphase of cell division with colchicines and controlling concentration and time, hypotonic solution time and trypsogen digesting time, we compared different effetcs of chromatosome-making. Our results shows we can make more adequate chromatosome specimen if the concentration of colchicines is 0.2μg/ml and the action time is 6h, the time of hypotonic solution is 35min and digesting time of trysogen is 20s, karyotype identification is normal diploid. So we can search for adequate Chromatosome Specimen-Making condition of MSCs and provide foundation for advanced study on MSCs.
