摘要
血液动力学主要是应用流体力学理论和方法研究血液流动、血管生理和病理之间关系的一门边缘性学科。近年来国内外学者认为血液动力学因素对血管重构有着重要的影响 ,当血液动力学发生改变后 ,血管内皮细胞通过跨膜蛋白、信号传导和基因表达等将力学信息传递到细胞内 ,再经过效应分子将转导信号最终作用于相应的血管 ,从而参与血管形态和功能的重构过程。本文旨在简述血液在流动过程中 ,血液动力学因素对血管内皮细胞的形态和功能、平滑肌细胞增殖和细胞外基质的生成等的影响。
Hemodynamic forces play an active role in many physiological and pathophysiological processes of the cardiovascular system. Vascular remodeling, changes of structure, geometry and mechanical properties of the artery, plays a role in many (patho)physiological processes. Several lines of evidence support a role for hemodynamic forces in the development and progression of vascular cell, such as endothelial cell, vascular smooth muscle cell and extracellular matrix. This review focuses on the role of hemodynamic forces in vascular remodeling relevant to vascular diseases. Cellular signal involves a complex interplay between cytoskeletal and biochemical elements and results in changes in G proteins, signal transduction and gene expression. We review current knowledge on flow-induced vascular remodeling and summarize the molecular mechanisms believed responsible for endothelial cell gene regulation with an emphasis on signal transduction.
