摘要 目的合成检测尿酶的两种新底物并建立对肾损伤动态监测方法。方法通过缩合及脱乙酰化分别合成CNP-NAG和CNP-Gal,并建立速率法分别测定庆大霉素肾损伤儿童尿中NAG酶及Gal酶活性及G/N的比值。结果所合成的试剂各项指标与进口样品完全一致。当检测因庆大霉素引起儿童肾损伤时,发现G/N的比值更能反映儿童肾损伤状况及肾功能修复的状况。结论可将肾功能指标由静态监测提升到动态监测,并确定G/N=0.15为肾损伤及肾修复的较敏感指标。 Objective To Synthesis two noval substrates for measuring urinary enzyme activities and establish dynamic method on monitoring kidney injury. Method by condensation and deacetylation reaction,CNP-NAG and CNP-Gal compounds was prepared. They are used for rate assay method.The value of NAGase, GALase and ratio G/N were detected respectively. Results The various parameters of two compounds are as same as the foreign products. When measuring kidney injury caused by gentamicin in children, it is found that the value of ratio G/N reflects the level of kidney injury and kidney repairment. Conclusion The detection level were elevated from static into dynamic monitoring,and the ratio G/N=0.15 was sensitive index for the renal assay.
