吸烟对人外周血单个核细胞NF-κB和IL-8表达的影响 被引量:2

摘要 目的 探讨吸烟对人外周血单个核细胞 (PBMC)NF κB活化和IL 8表达的影响。方法 选择健康吸烟者和非吸烟者各 8名 ,观察核因子 кB (NF кB)胞核染色阳性细胞百分比、I кB蛋白、白细胞介素 8(IL 8m)RNA及细胞上清IL 8浓度的变化。结果 未经LPS处理的吸烟者PBMC的NF кB染色阳性细胞百分比、I кB蛋白、IL 8mRNA及细胞上清IL 8浓度均分别显著高于未经LPS处理的非吸烟者 (P <0 .0 5 )。以 10 μg/L的LPS刺激后 ,上述指标在非吸烟者中无明显变化 (P >0 .0 5 ) ,而在吸烟者中则显著升高 (P <0 .0 1)。结论 脂多糖刺激吸烟者外周血单个核细胞NF кB活化增强 ,IL 8释放增多 。 Objective To explore the effect of smoking on nuclear factor кB and interleukin 8 in human peripheral blood mononuclear cells(PBMCs).Methods PBMCs were collected from 8 heathy smokers and 8 heathy nonsmokers.The NF кB activity in PBMCs,IL 8 mRNA and the concentration of IL-8 in supernatant were measured by immunocytochemical staining,western blot,RT PCR and radioimmunoassay.Results Compared with nonsmokers,the percentage of nuclear positive cells of NF кB,I кB in cytoplasm protein,IL 8 mRNA and IL 8 protein in supernatant were higher in smokers( P <0.01).By pretreatment with 10μg/L LPS,the above index had no effect in nonsmokers( P >0.05),but there were a significantly increase in smokers( P <0.01).Conclusion The difference in the activation of NF кB and concentration of IL 8 in smokers and nonsmokers may related to chronic exposure of PBMCs to components of smoke.It may contribute to the increased susceptibility of smokers to baterial infections.
