摘要
目的 观察Survivin在星形细胞瘤中的表达 ,通过分析其与VEGF和微血管密度的相关性 ,探讨其与肿瘤血管形成的关系。方法 星形细胞瘤手术标本 35例 ,免疫组化方法检测肿瘤组织中的Survivin ,用CD34单抗显示微血管 ,用血管计数 (MVD)定量肿瘤微血管形成。RT -PCR检测肿瘤组织中VEGFmRNA的表达。结果 1Survivin、VEGFmRNA和MVD在星形细胞瘤中的表达与肿瘤的病理分级呈正相关 (P <0 .0 1)。 2在Survivin阳性标本中VEGFmRNA和MVD的表达均高于Survivin阴性的标本 ,差异有显著性 (P <0 .0 1P <0 .0 5 )。 3Survivin的表达强度与VEGFmRNA的表达行线性回归分析 ,呈显著正相关 ,R =0 .814 ,P <0 .0 1。结论 Survivin的表达随星形细胞瘤的恶性程度升高而增高 ,说明它是肿瘤恶性的特征。Survivin的表达与VEGF和MVD的表达呈正相关 。
Purpose: To observe the expression of Survivin in astrocytoma of various pathological grades and explore its relation to tumor vascularization. Methods: Thirty-five astrocytoma samples were obtained from the patients who underwent neurological surgery. Survivin was stained by way of immunohistochemistry and the microvessel was immunostained with anti-human CD34 mono-antibody. MVD was measured to show tumor vascularization as well as IOD of Survivin positive cells being checked. Semi quantitative RT-FCR was also applied to detect VEGF mRNA expression in astrocytoma. Results: The expression of Survivin, VEGF mRNA and MDV were significant positive correlated with the pathological grades of astrocytoma (P<0.01). Expression of VEGF mRNA and MVD in the Survivin positive group were considerably higher than that of the Survivin negative group (P< 0.01 and <0.05 respectively). In the Survivin positive group, the Sruvivin expression is significantly positive correlated with VEGF mRNA by the linear regression analysis (R=0.814, P<0.01). Conclusions: Survivin tended to express in high-grade ascrocytoma, which suggested that it could be a mark of malignant tumor. And that, the expression of survivin went up with VEGF and MVD led to the possibility that Survivin may take park in the VEGF-related tumor angiogenesis.
