摘要
本文用乙酰葡萄糖异硫氰酸酯(GITC)作柱前手性衍生化试剂,反相高效液相色谱法拆分氨基酸立体异构体。研究了不同构型氨基酸与GITC反应后所生成的衍生物的紫外吸收响应特点。探讨了16种氨基酸GITC衍生物的色谱分离情况。
A reversed-phase high-performance liquid chromatographic method for the separation of amino acid stereo-isomers was developed, In the method, 2, 3, 4, 6-tetra -O-acetyl-β-D-glucopyranosyl isothio-cyanate was chosen as the reagent for the pre-column derivatization of enantiomeric amino acids to give di-astereomeric thiourea derivatives. These derivatives were efficiently separated on a C18 column. The ultraviolet absorption response of both the D-and L-amino acid derivatives has been investigated. The resolution of 16 amino acids has been presented.
