雌激素样作用中药复方对卵巢切除大鼠雌激素受体和β-内啡肽的影响 被引量:14

Effects of estrogen-like herb extracts on estrogen receptors and β-endorphin in ovariectomized rats

摘要 目的 探讨以补骨脂和小茴香为主的雌激素样作用中药复方 (CHF)对卵巢切除大鼠雌激素受体 (ERs)和 β 内啡肽 (β EP)的影响。 方法 体质量 2 60~ 2 80g雌性Wistar大鼠 ,卵巢切除 (OVX)后 4周 ,每天分别灌胃给予 5、10、2 0g kg的CHF提取物 ,同时设卵巢切除对照、雌激素对照 (OVX ,0 .1g kg 17β 雌二醇 )和假手术对照 (SHAM)。测定血清雌二醇(E2 )、黄体生成素 (LH)、下丘脑视前区 (HPO)和血浆的 β EP浓度以及HPO的ERs含量。 结果  5、10、2 0g kg的CHF提取物均可显著升高卵巢切除大鼠的E2 浓度并降低LH浓度 ,显著增加HPO的ERs含量 (P <0 .0 5 ) ;而CHF提取物可降低HPO的 β EP含量 ,并呈剂量反应关系 ;但 2 0g kg的CHF提取物可显著升高血浆 β EP浓度 (P <0 .0 5 )。 结论 CHF提取物与 17β 雌二醇相似 ,可改善由卵巢切除造成的ERs异常和 β Objective To examine the effects of estrogen like herb extracts mainly containing Fructus Foeniculi and Fructus Psoraleae (CHF) on estrogen receptors (ERs) and β endorphin (β EP) in ovariectomized (OVX) rats. Methods Female Wistar rats weighing 260-280 g were bilaterally ovariectomized under anesthesia. After 4 week recovery, CHF at the doses of 5, 10, and 20 g/kg was administered by gavage for 12 weeks. The positive group (17β estradiol, 0.1 g·kg·d -1 ), sham group and OVX control group were also included simultaneously in present study. Serum estradiol (E2) concentrations were determined at 5, 8, 12, and 16 weeks. ERs contents in hypothalamus preoptic area (HPO) andβ EP concentrations in HPO and plasma were also measured at week 16. Results OVX rats in groups treated with CHF at the doses of 5, 10, and 20 g·kg·d -1 had significantly higher E 2 and lower LH concentrations and HPO ERs contents than animals in OVX control group ( P <0.05). CHF decreased β EP in HPO in a dose dependent manner. Rats treated with CHF at the dose of 20.0 g·kg·d -1 increased β EP content as compared with animals in OVX control group ( P <0.05). Conclusion Similar to 17β estradiol, CHF could prevent against the disturbance of ERs and β EP induced by estrogen deficiency in OVX rat models.
