摘要
目的 :探讨中药方剂在体内动态过程对药效影响的规律。方法 :小鼠分别 ig五味子乙素和五灵胶囊 ,在不同时间点采集血浆 ,加硫酸胺与乙腈 -甲醇 (4∶ 1)混合液提取制备样品液 ,经 HPL C分析乙素含量。结果 :小鼠 ig给药后 (含乙素4 5 .0 mg/ kg) ,五灵胶囊所含乙素的药代动力学模型为单室模型一级吸收 ,其药代动力学参数在小鼠胃内 K a=1.5 6 1 /h、Ke=0 .5 11 /h,TMax=1.0 5 h,T1 /2 ke=1.33h;供试品乙素 (4 5 .0 mg/ kg) Ka=1.891 /h,Ke=0 .2 5 1 /h,TMax=1.2 5 h、T1 /2 ke=2 .71h;在肠内 Ka=14 .6 3h,Ke=0 .2 5 h,T( Max) =3.6 5 h,T1 /2 ke=2 .71h。结论 :中药方剂改变了中药所含化学成分在体内生物药剂学特征 ,使其在体内药效作用延长 。
Objective:To investigated the law of Chinese traditional medicine formula in vivo dynamic course effects to the pharmacodynamic.Methods:The schizandrin and Wuling capsules were given to mice individualy by irrigate stomach, the blood plasma was collected in dissimilarity time, the sample fluid was prepared to extract adding amine sulfate and mix fluid of Acetonitrile-methyl alcohol (4∶1), and which to be detected content of schizandrin by HPLC.Results:After irrigated the drugs to mice, the plasma concentration-time course fitted well to one-compartment model with the 1st order absorption and with the following pharmacokinetic parameters:the parameter of schizandrin were Ka=1.56 1/h,Ke= 0.51 1/hT Max= 1.05 h,T 1/2ke=1.33 h; the parameter schizandrin of Wuling capsules in stomach were Ka=1.89 1/h,Ke=0.25 1/h,T Max=1.25 h,T 1/2ke=2.71 h; those in intestinal were Ka=14.63 h,Ke=0.25 h,T (Max)=3.65 h,T 1/2ke=2.71 h.Conclusion:The formula of Chinese traditional medicine changes the character of biopharmaceutics of mostly chemical components in Chinese medicine in vivo. The effect of pharmacodynamic prolongs and the bioavailability increases.
