摘要
目的:观察李氏5号方水提液对D-半乳糖老化小鼠海马神经元凋亡级联反应的影响。方法:将小鼠随机分为5组:正常对照组(C组)、D-半乳糖模型组(D组)、李氏5号方大剂量药物治疗组(L组)、中剂量药物治疗组(M组)、小剂量药物治疗组(S组)。D,L,M,S4组每日皮下注射半乳糖65mg/kg。L,M,S3个治疗组分别灌胃李氏5号方0.6,0.3和0.15g/kg,连续3个月。免疫组织化学染色方法和WesternBlotting检测海马神经元(凋亡诱导因子(AIF),Bax细胞色素C(CytC),Bcl-2的表达。结果:模型小鼠海马神经元AIF,Bax,CytC的阳性细胞数分别比正常对照小鼠增加4.4倍、6.2倍和4.4倍,而Bcl-2则仅为正常小鼠的12%。李氏5号方水提液3种不同剂量均不同程度使这4种蛋白质表达的异常有所恢复,但未能达到完全正常水平。结论:D-半乳糖老化小鼠的海马神经元存在明显的凋亡级联反应,李氏5号方水提液对改善凋亡级联反应有一定作用。
AIM:To observe effect of Lishi No.5 formula liquid extract on the apoptosis ca scade reaction of hippocampus neuron in aging rats induced by D-galactose. METHODS:Rats were randomly divided into 5 groups:control group(group C),D-gal actose model group(group D), large-dose Lishi No.5 formular liquid extract trea tment group(group L) ,moderate-dose treatment group(group M),and small-dose tr eatment group(group S).Rats in groups D,L, M, and S were injected galactose(65 m g/kg)subcutaneously every day,while rats in group L,M,and S were filled into sto mach with Lishi No.5 for 3 months(0.6 g/kg, 0.3 g/kg, 0.15 g/kg,respectively).T he expressions of apoptosis inducing factor (AIF)of hippocampus neuron,Bax,cytoc hrome C(CytC),and Bcl-2 were tested with immunohistochemical staining and Weste rn blotting. RESULTS:The number of positive cells of AIF,Bax,and Cyt C in the hippocampus o f model rats increased to 4.4, 6.2, 4.4 times more than that of the control grou p respectively,while the number of Bcl-2 was only 12% of that of normal rats .Lishi No.5 formular liquid extract with different dosages promoted the rehabili tation of the abnormal expressions of the 4 proteins to some extent, but not rec overed to the normal. CONCLUSION:The apoptosis cascade reaction of hippocampus neurons in aging rats induced by D-galactose is obvious.Lishi No.5 formular liquid extract plays a r ole in improving the apoptosis cascade reaction.
