摘要
目的 :建立高效液相色谱测定兔房水中丹参素含量的方法 ,了解兔眼丹参注射液直流电离子导入后房水中丹参素的吸收情况。方法 :色谱条件 :色谱柱为HypersilDivisionODSC1 8分析柱 (1 5 0mm× 4 .6mm ,5 μm) ;流动相为水 甲醇 (95∶5 ) ,1 0 0 0mL中含十二烷基磺酸钠 1 g ,磷酸二氢钠0 .6 g,冰醋酸 2mL ;流速为 1 .0mL·min-1 ;检测波长为 2 81nm ;柱温为 35℃。结果 :在 0 .5 2~ 8.32mg·L-1 范围内 ,峰面积与浓度呈良好线性关系 ,r =0 .9998,最低检测浓度为 0 .0 5mg·L-1 ;平均回收率为 93.4 2 %;日内和日间RSD均 <1 0 %。结论 :建立了特异 ,简便 ,灵敏的HPLC检测法 ,为进一步研究探讨丹参素在眼内作用的机理打下基础。
OBJECTIVE To detemine danshensu in rabbit aqueous humor with HPLC and to observe the effect of direct current on transmitting the ion of Danshensu.METHODS The analytical column was a ODS C 18 stainless steel column (150 mm× 4.6 mm, 5 μm); water∶methanol(95∶5) (patassium biphosphate 0.6 g, dodecyl sodium sulfate 1.0 g and Acetic acid 2 mL in 1000 mL mixture) as mobile phase, flow rate was 1.0 mL·min -1 and detection wavelength was 281 nm. The column temperature was 35℃. RESULTS The relationship between the peak area and concentration of the drug is linear within the range of 0.52~ 8.32 mg·L -1 with a correlation coefficent of 0.9998.The detection limit was 0.05 mg·L -1. The average recovery was 93.417%;RSD of intra-day and inter-day was less than 10%. CONCLUSION A specific, simple, sensitive and suitable method is established for the research of mechanism of Danshensu in ophthalmological diseases.
