摘要
目的 探讨美多芭对帕金森病大鼠细胞凋亡的影响。 方法 采用 6 羟基多巴胺(6 OHDA)注射于大鼠脑右侧黑质造成偏侧帕金森病模型 ,并用美多芭进行治疗 ,同时设立正常对照组、假手术组。用TUNEL染色和透射电镜的方法观察美多芭对神经细胞凋亡的影响。 结果 美多芭治疗组TUNEL染色阳性细胞 (2 3 5 0± 3 85 )个 ,比模型组 (37 83± 6 4 6 )个明显减少 ,阳性细胞计数结果比较 ,两组差异有显著性 (P <0 0 1)。电镜检查结果显示美多芭治疗组细胞改变明显比模型组为轻 ,表现为细胞膜稍有皱缩 ,细胞体积略缩小 ,线粒体等细胞器形态正常 ,细胞核稍呈皱缩状态 ,染色质有轻度凝聚成块并有边聚倾向 ,核膜基本完整。 结论 美多芭对帕金森病大鼠神经元细胞凋亡有明显抑制作用。
Objective To illustrate the effect of Madopar on nerve cell apoptosis of rats with Parkinson’s disease (PD). Methods Hemi-lateral PD model was made by injecting 6-OHDA into the black substance of the right side of the brain and treated with Madopar. At the same time,the normal group and sham-operation group were set up,and TUNEL staining and transmission electro-speculum were employed to observe the apoptosis of nerve cells. Results The TUNEL positive cells(23.50±3.85) were significantly less than the model group(37.83±6.46) and the comparison of apoptosis cells counting showed marked difference( P <0.01). The electro-speculum examination showed that the pathological changes of the treated group were milder than the model group;the cell membrane appeared a bit shrinking,the cell volume appeared a little smaller than the baseline,the cell organ form including thread granules appeared normal,the cell nucleus appeared a bit shrinking,the chromatin showed agglutination with tendency of border agglutination,and the nucleus membrane kept basically complete. Conclusions Madopar has a significant inhibitory effect on the apoptosis of neuron cells of rats with PD.
