摘要
目的 :探讨药理浓度的三氧化二砷 (As2 O3 )对雄性大鼠血清睾丸酶及精子生成的影响。方法 :不同剂量的As2 O3 (2mg·kg-1·d-1,4mg·kg-1·d-1和 8mg·kg-1·d-1)给予大鼠腹腔内注射 ,2周后检测大鼠血清睾丸酶的含量 ,同时 ,测定大鼠血清砷的含量、睾丸脏器系数、睾丸精子头计数 ,并计算每日精子生成量。结果 :中剂量组各种酶都有所下降 ,G 6 PD、LDH及LDH X的减少有统计学意义 (P <0 .0 1,P <0 .0 1和P <0 .0 5 ) ;大剂量组ACP、ALP、G 6 PD、LDH及LDH X均明显下降(P <0 .0 1)。随着给大鼠腹腔注射的As2 O3 浓度增加 ,血中砷浓度明显增加 (P <0 .0 1)。 3种剂量组的睾丸脏器系数与对照组差异均无显著性 (P >0 .0 5 ) ,大剂量组睾丸精子头数和每克睾丸每日精子生成量与对照组比较减少明显 (P <0 .0 1) ,血清砷含量与大鼠精子头数之间呈直线负相关 (r =- 0 .5 15 ,P <0 .0 1)。结论 :As2 O3 对睾丸酶的抑制 。
OBJECTIVE To explore the effects of pharmacological concentration of arsenic (As 2O 3) on enzymes of testis and sperm production in male rats.METHODS Different doses of As 2O 3 ( 2 mg·kg -1·d -1, 4 mg·kg -1 ·d -1 and 8 mg·kg -1·d -1) were administrated ip to the adu lt male rats.After two weeks,the levels of enzymes in testis were observed,serum contents of arsenic were determined;the coefficient of testicular viscera were counted;the numbers of testicular sperm head were counted and daily sperm produ ction was calculated.RESULTS The levels of glucose-6-phosphatases-dehydrogenase(G-6-PD),lactate dehydrog enase isozyme (LDH) and lactate dehydrogenase isozyme-X(LDH-X) of testis in me dium dose group decreased significantly(P< 0.01,< 0.01 and < 0.05) .In large dose group,the contents of acid phosphatases (ACP),alkaline phosphatas es (ALP),G-6-PD,LDH and LDH-X were lower than that of controls (P< 0.05 ~ 0.01 ).There were no differences in the coefficient of testicular viscera in three groups.The number of testicular sperm head and daily sperm production in large- doses group decreased significantly than that of control (P< 0.01).There were negative correlation between the content of serum arsenic and the sperm he ad counts(r= -0.515,P< 0.01).CONCLUSION By means of inhibiting enzymes of testis,restraint of sperm production in male r ats,As 2O 3 may give rise to male reproduction toxicity.
