摘要
目的 :观察槲皮素对氚标记的血小板活化因子与血小板膜上受体结合的影响 ,试图证明该药为一新型血小板激活因子 (PAF)受体拮抗剂。方法 :以放射配基结合试验观察 [3H]PAF与兔血小板受体特异性结合的作用 ;以分光光度法测定PAF诱发血小板黏附的强度。结果 :槲皮素可浓度依赖地抑制2 5 ,5 0 ,10 0nmol·L- 1 [3H]PAF与兔血小板受体的特异性结合 ;该药可明显抑制PAF诱发的血小板黏附 ,具明显的量效关系 ,其IC50 为 39 8μmol·L- 1 。结论 :槲皮素具抗PAF作用 ,为一新的PAF受体拮抗剂。
Objective: To observe the platelet activating factor (PAF) receptor binding antagonistic effect of Quercetin. Method: The specific binding of [ 3H]PAF to rabbit platelet receptor was investigated by radio ligand binding assay (RLBA). Platelet adhesion induced by PAF was determinated by spectrophotometry. Result:The specific binding inhibition effect of Quercetin was found to be concentration-dependent in [ 3H] PAF 2.5, 5.0, 10.0 nmol·L -1 . The washed rabbit platelet adhesion induced by PAF was inhibited by Quercetin in a dose-dependent manner. The IC 50 of Quercetin was 39.8 μmol·L -1 . Conclusion:The specific receptor binding of PAF can be antagonized by Quercetin.
