摘要 目的:观察野生型p53基因转染对前列腺癌细胞PC鄄3m增殖和凋亡的影响。方法:用脂质体Lipofectamine将真核表达载体pCMV鄄p53和pCMV鄄p53mt135分别转染PC鄄3m细胞,MTT法检测转染前后细胞生长情况,转染48h后采用流式细胞技术(FCM)检测细胞凋亡率。结果:pCMV鄄p53转染后的PC鄄3m细胞生长明显受到抑制,FCM显示,转染48h后,细胞凋亡率为15%。结论:野生型p53基因瞬间转染PC鄄3m细胞可抑制细胞增殖,诱导细胞凋亡。 Objective: To observe the effect of transient transfection of wild-type p53 gene on the proliferation and apoptosis of prostate cancer cell line PC-3m. Methods: The eukaryotic expression vector pCMV-p53 was transfeced into PC-3m cells with lipofectamine. The cell growth was observed before and after transfection by MTT assay. The cells were collected 48 hours after transfection for determining apoptotic rate by flow cytometric (FCM) analysis. Results: The PC-3m cell growth after transfection was inhibited by MTT. The apoptotic rate was 15% 48 hours after transfection. Conclusion: The transient transfection of wild-type p53 gene induces the apoptosis of prostate cancer cell line PC-3m.
