摘要
目的 观察牛磺酸对系膜细胞增生及细胞外基质分泌的影响 ,以探讨其作用的可能机制。方法 用MTT法检测系膜细胞增生 ,流式细胞仪检测细胞周期 ,ELISA检测细胞外基质Ⅳ型胶原的生成。结果 牛磺酸可剂量依赖性地抑制系膜细胞增生 ,并影响细胞周期 ,使G0 /G1期细胞增多 ,S期细胞减少 ,并可明显减少Ⅳ型胶原的分泌 (P <0 .0 1)。结论 牛磺酸可显著抑制系膜细胞增生 ,使细胞周期阻滞于G0 ~G1期 。
Objective To investigate the effect of Taurine on proliferation and extracellular matrix secretion of rat mesangial cells in vitro. Method Rat mesangial cells were incubated with different concentration of taurine.Cell proliferation was assessed by MTT colorimetric assay.Cell cycle was detected by flow cytometry,and type Ⅳ collagen was determined by ELISA. Results Administration of 5~80 mmol/L taurine into culture medium had no toxicity to mesangial cells,but could significantly inhibit mesangial cells proliferation in dose-dependent manner,subsequently increased the cells in G 0/G 1 phase and decreased the cells in S phase.In addition,Taurine could markedly inhibit type Ⅳ collagen and laminin secretion in mesangial cells ( P <0.01). Conclusion Taurine can significantly inhibit mesangial cell proliferation and type Ⅳ collagen secretion.
