摘要
目的 :检测链脲佐菌素 (STZ)诱导的糖尿病 (DM)大鼠早期肾脏中肝细胞生长因子 (HGF)和P2 1的表达 ,探讨HGF在DM大鼠早期肾小球肥大中的作用。方法 :STZ诱导DM大鼠动物模型后第 3,7,14d ,分别用免疫组化法、ELISA法和RT PCR检测了肾脏中HGF蛋白及mRNA的表达 ,同时用免疫组化法检测了肾脏中P2 1蛋白的表达。结果 :STZ诱导的DM大鼠早期 ,HGF在肾脏中的表达主要位于肾小球内 ,HGF蛋白和mRNA均于STZ注射后的第 3d开始升高 ,并于第 7d时达到高峰 ;肾小球内P2 1蛋白的表达于STZ注射后的第 7d达到高峰 ;DM大鼠肾小球内HGF蛋白水平和P2 1蛋白积分呈正相关 (r=0 .9346 ,P <0 .0 1)。结论 :STZ诱导的DM大鼠早期肾小球内有HGF的表达 ,其表达可能与DM早期肾小球肥大有关。
Objective: To evaluate the glomerular expressions of hepatocyte growth factor (HGF) and P21 in early streptozotocin-induced diabetic rats and to explore the role of HGF in glomerular hypertrophy. Methods: The protein expressions of HGF and P21 were measured by immunohistochemisty at days 3, 7 and 14 after STZ administration. The glomerular HGF protein and mRNA level was determined by sandwich enzyme-linked immunosorbent assay (ELISA) and reverse transcription-polymerase chain reaction (RT-PCR) respectively. Results: The expression of HGF was mainly detected in glomeruli and reached a peak on day 7. The protein expression of P21 also reached a peak on day 7. There was a positive correlation between HGF protein level and P21 protein score (r=0.9346, P<0.01) in glomeruli of diabetic rats. Conclusion: The protein and mRNA expression of HGF is expressed in glomeruli in early streptozotocin-induced diabetic rats and it may play a role in the early diabetic renal hypertrophy.
