研究了在相同培养条件下不同培养液、不同血清和不同细胞密度对绵羊睾丸细胞生长的影响 ,并对该细胞接种羊痘弱毒后的病变情况进行了观察。结果 ,3种培养液对绵羊睾丸细胞生长的支持作用表现为MEM液 >199液 >乳汉液 ,但差异不显著 ;用犊牛血清的最适细胞密度为8.0× 10 5~ 1.0× 10 6 /mL ,用成年牛血清的最适细胞密度为 1.0× 10 6 /mL ,用绵羊血清的最适细胞密度为 8.0× 10 5/mL ;
Effect of cell culture media, sera and cell density on growth of ovine testicular cells was studied under the identical culturing condition and pathological changes of the cells inoculated with the (attenuated) sheep pox virus was observed. The results were as follows. The support effect of MEM, 199 and milk-Hank’s solutions on growth in vitro of the cells was obvious (in support order: MEM>199>milk-Hank’s (solution)). The most suitable densities of the cells in calf serum, mature bovine serum and (ovine) serum were 8.0×10~5-1.0×10~6/mL, 1.0×10~6/mL and 8.0×10~5/mL respectively. The grown-well ovine testicular cells appeared evident pathological changes after they were innoculated with the attenuated sheep pox virus.
