摘要
目的 研究生长激素对全脑缺血 /再灌注后大鼠脑神经细胞的保护作用。方法 SD大鼠 18只 ,雌雄各半 ,随机分 3组 :对照组、CPR 2 4h组及生长激素组 ,每组 6只。采用窒息合并冰氯化钾停跳液致大鼠心跳骤停 -心肺复苏模型 ,停跳 5min后开始CPR ,采用免疫组织细胞化学方法观察各组海马区神经元中bcl- 2和bax的表达情况。结果 生长激素组bcl- 2表达明显较常规复苏组和对照组高 ,而TUNEL阳性细胞少。结论 生长激素可能促进复苏早期神经元表达抗凋亡蛋白bcl- 2 ,抑制神经元的凋亡 。
Objective To investigate protective effect of recombinant human growth hormone on cerebral ischemia-reperfusion in rats. Methods Cardiac arrest was induced by asphyxiation and ice-cold 0.5M KCl in rats and resuscitation was begun five minutes after arrest. Sprague Dawley rats 18, half female and half male, were randomly divided into 3 groups: control group (n=6); 24-hour cardiopulmonary resuscitation group(n=6);recombinant human growth hormone group(n=6). Bcl-2 and bax protein expression in the rat brain were studied using immunocytochemistry after resuscitation. Result Expression bcl-2 was higher and TUNEL-positive cells were less in growth hormone group than other groups. Conclusion Recombinant human growth hormone might promote expression of bcl-2, restrain neuron apoptosis, alleviate hippocampa injury of the rats.
