摘要
目的 利用水溶性甲簪显色法测定山梨醇脱氢酶的活性。以此作为肝功能的特异性指标。方法 对 32 5例正常成人和 82例肝病患者的血清与自研试剂作用后生成紫色化合物———甲簪 ,在CL 72 0 0型生化分析仪上测定其酶的活性 ,并对甲簪试剂进行一系列可行性分析。结果 甲簪试剂最大吸收峰波长为 5 72 6nm ,其酶促反应最适pH为 8 8,线性敏感度为 6 0 μmol L ,并且有一定的抗干扰能力和一定的稳定性。正常人血清中山梨醇脱氢酶活性为 2 4 3± 1 0 4U L ,肝病患者血清山梨醇脱氢酶活性为 6 74± 3 10U L。结论 肝病组山梨醇脱氢酶活性明显高于正常组 ,可作为临床观察肝脏功能的一项特异性指标。甲簪显色法用可见光波长在 5 70nm比色有较好的线性关系和稳定性 。
Objective To convert foramzan to be water soluble, and measure the activity of SDH with the foramzan using chemical solubilizer. Methods The serum of 325 normal adults and 82 patients with hepatic disease react with a series of reagents to form foramzan. The foramzan detemined was CL 7200 biochemical analyzer. And a lot of feasibility analysis of foramzan reagent was made. Results the activity of SDH in the serum of normal group was 2 43±1 04 U/L, the activity of SDH in the serum of patient was 6 74±3 10 U/L, The maximum absorption of Foramzan was at 572 6 nm. the optimal pH of the enzyme reaction was 8 8, the linear sensitivity was 60 umol/l, and has very high stabilization and anti jamming. Conclusion The SDH in patients was significantly higher than the SDH in normal adult, the solubilizer works very well in the reaction. The fromzan regent has very stabilization and anti jamming, can be used in a series of measurement of enzyme activity which need NAN(P)H coenzyme.
