U50,488H预处理及低温保存离体兔心不同时限超微结构的变化

摘要 目的探讨U50,488H预处理及低温保存对离体兔心的保存效果和不同保存时间对心肌超微结构的影响。方法健康大白兔32只,随机分成4组,每组8只。1组:低温保存4 h;2组:低温保存6 h;3组:低温保存8 h;4组:低温保存10 h。每组动物经50U,488H预处理后,离体供心均用U50,488H心肌保存液进行低温保存。用电镜观察保存后的心室肌超微结构,比较不同保存时间心肌超微结构的变化。结果兔心保存4 h和6 h时心肌细胞出现轻度损伤;在保存8 h和10 h心肌细胞呈现中度损伤。但保存10h的离体心脏再灌注后心功能的恢复与保存8 h组相比较存在明显的差异。结论U50,488H药物处理及心脏低温保存供心8 h和10 h后心肌组织均出现中度损伤,但离体兔心保存10 h再灌注后出现严重的心功能障碍,而兔心保存8 h以内的离体心脏保存效果良好。 Objective: To investigate the protective effect and ultrastructure changing of rat′s heart by U50, 488H preconditioning and U50, 488H heart preservation solution to storage, in hypothermia for different preservation time. Methods: Healthy rabbits (n=32) were randomly divided into four groups, eight rabbits in every groups. Group one, the isolated rabbits′ hearts were preserved for hours in hypothermia; group two, for six hours; group three, for eight hours; group four, for ten hours long. All rabbits′ were pretreated by U50, 488H and isolated rat heart be preserved by U50, 488H preservation solution in low temperature (0~4)℃. To compare the different change among the rat′s myocardium ultrastructure in different time. Results: When protection the heart for four hours and six hours, the cardiac muscle cell showed up minor injury; the cardiac muscle cell showed up secondary level in 8 h group and 10 h group. After reperfusion, the recovery degree of heats function, there were obviously different in 8 h group and 10 h group. Conclusion: The myocardium tissue appears secondary level injury in 8 h and 10 h by U50, 488H preconditioning and heart preservation solution preserve in hypothermic, but there are serious heart function disturbance in 10h group, the whole preservative result is fine in 8h group.
