摘要
目的:研究胰岛素样生长因子Ⅰ(IGF-1)对大鼠皮肤创面愈合过程的影响。方法:以大鼠为动物模型,在伤后1,4,7,11,14,和17d取材,利用免疫组织化学和图像分析,研究皮肤创面自然愈合过程中创面IGF-1前体、内源性IGF-1和增殖细胞核抗原(PCNA)的动态变化,以及它们与创面愈合相关因素成纤维细胞之间的关系。结果:在皮肤创面自然愈合过程中,内源性IGF-1表达量的变化与成纤维细胞活动、PCNA表达的变化基本相符,与IGF-1前体的变化关系不密切。结论:创面愈合过程中,在局部发挥生理效应的IGF-1主要来自创面以外的来源。内源性IGF-1与成纤维细胞增殖、活跃程度密切相关。
AIM:To study the effect of type I insulin like growth factor (IGF 1 ) in the process of wound healing on the back skin of rat. METHODS: The wound models were made on the back skin of rats, then the specimens in the wound site were harvested after 1, 4, 7, 11,14 and 17 days.Immunohistochemical method and image analysis were used to detect the changes of endogenous IGF 1, proIGF 1 and proliferating cell nuclear antigen(PCNA) during wound healing.The relationship among proIGF 1, IGF 1 and their related factor of wound healing and fibroblast were investigated. RESULTS:The changes of endogenous IGF 1 in the process of wound healing mainly coincided with the changes of fibroblast and the expression of PCNA, but not closely related with the changes of proIGF 1. CONCLUSION: In the process of wound healing, IGF 1 that locally plays physiological effect mainly comes from outer source of the wound. Entogenous IGF 1 has a closely relation with the proliferation and the active degree of fibroblast.
