摘要
背景:有证据表明,大血管内皮细胞对高血糖症具有与视网膜微血管不同的特征与反应。目的:检验大剂量葡萄糖水平对培养的原代人视网膜毛细血管内皮细胞(humanretinalcapillaryendothelialcells,HRCECs)的直接影响。设计:非随机非对照的实验研究。地点和对象:本研究在解放军第四七六医院内科完成。对象为HRCEC细胞,从志愿捐赠者眼中分离得到。方法:HRCEC细胞在含有5mmol/L或25mmol/L葡萄糖的培养基中培养6d。应用锥虫蓝染色测定细胞活力、流式细胞计数分析细胞周期、TUNEL分析细胞凋亡。主要观察指标:在高剂量葡萄糖中HRCEC的增殖和凋亡的量化。结果:HRCEC细胞在25mmol/L葡萄糖中培养6d,细胞活力显著下降。通过流式细胞计数和TUNEL测定,凋亡细胞的数目在大剂量葡萄糖培养基中显著升高。结论:大剂量葡萄糖可诱导人视网膜血管内皮细胞凋亡,这有可能是促进糖尿病性视网膜病发展的原因。
BACKGROUND:It is proved that the macrovascular endothelial cells differ from those of retinal microvascular cells in their features and response to hyperglycemia. OBJECTIVE:To examine the direct effect of high level glucose on primarily cultured human retinal capillary endothelial cells(HRCECs). DESIGN:A non randomized uncontrolled experimental study was performed. SETTING and PARTICIPANTS:The experiment was conducted in the Department of Internal Medicine,the 476 Hospital of Chinese PLA.The subjects were HRCECs isolated from eyes of volunteer donors. INTERVENTIONS:HRCECs were cultured in media containing 5 mmol/L or 25 mmol/L glucose for 6 days.The cell vitality was examined with trypan blue exclusion assay,the cell cycle was analyzed with flow cytometry,and the cellular apoptosis was assayed with TUNEL method. MAIN OUTCOME MEASURES:The quantified proliferation and apoptosis of HRCECs in high dose glucose. RESULTS:The vitality of cells decreased significantly after exposure to 25 mmol/L glucose for 6 days.The number of apoptotic cells was determined with flow cytometry and TUNEL increased significantly in response to high dose glucose treatment. CONCLUSION:High dose glucose induces apoptosis of HRCECs,which may contribute to the development of diabetic retinopathy.
