摘要
目的:优选栀子药材的提取工艺。方法:采用直接比较法、正交试验设计法,以HPLC法测定各样品中栀子苷的含量为指标,比较了提取方法和提取溶媒种类、溶媒用量、提取时间、提取次数对栀子苷含量的影响,并比较不同条件下的醇沉工艺。结果:以10倍量水,提取3次,每次提取1.5h为最优工艺,醇沉条件为提取液浓缩至相对密度为1.12(60℃),醇沉浓度为60%。结论:该工艺稳定、可行。
Objective:To optimize the process for extraction of Gardenia . Methods:The selection of solvent, quantity of solvent used, time and numbers of extraction were optimized by direct comparison and orthogonal design and gardenoside content in the extract was adopted to measure the superiority of the extraction process. HPLC was used to determine the gardenoside content in the extract. The alcoholic concentration suitable for precipitation and the relative density of extract were also optimized. Results:The optimal condition for extraction was as follows: the weight of water was 10 times of Gardenia ;the numbers of extraction was 3; and the time of extraction was 1.5h. The optimal condition for alcoholic precipitation was that relative density of the extract was 1.12 (60℃) and alcoholic concentration was 60% . Conclusion:The extraction process is suitable for practical use.
