摘要
本文旨在观察氧化修饰低密度脂蛋白(OX-LDL)对平滑肌细胞(SMCs)形态学的影响,探讨其在动脉粥样硬化形成中的意义.作者用硫酸铜诱导形成OX-LDL,以不同浓度加入培养传至第三代的正常家兔主动脉SMCs中,定期孵育后进行透射电镜观察,并以同样浓度的正常LDL为对照.与对照组比较,OX-LDL有更强的细胞毒性作用,随着浓度的增加,SMCs相继出现基膜及膜下小泡消失,肌丝结构模糊,密斑减少,肌浆网显著扩张,线粒体肿胀,线粒体脊结构消失及空泡状结构等病理变化.结果表明,OX-LDL对培养的动脉SMCs有毒性损伤作用,呈剂量—效应关系.
To investigate the significance of smooth muscle cells (SMCs) injured by oxidized low density lipoprotien (LDL) in the course of atherosclerosis. Coper-oxidized LDL was prepared by incubating LDL (100 fig/ml) with 5 μmol/L Cu2+in MEM medium at 37 C for 20h. Oxidized LDL and native LDL (50,100,200μg/ml respectively) were added to the dishes of cultured SMCs of the third generation. After 24h of incubation the cells were observed by electron microscopy. Comparing with the control group, Oxidezed LDL group was found to have severe cytotoxicity to SMCs. Disappearance of basement membrane and hypolemmal scrobicula, obscure of myofilament, diminution of dense patch, enlargment of endoplasmic reiticula, swollen mitochondria and bubble structure occurred seriously accompanied by increased concentration of oxidized LDL. The injury effects of oxidized LDL on the cultured SMC are stronger than those of native LDL. which may interact with atherogenesis.
