摘要
目的 比较弓形虫不同地理株 (RH株、ZS2 株、GT株 )GRA7基因的异同。方法 从弓形虫不同地理株的基因组DNA中扩增出GRA7基因 ,对目的基因用限制性内切酶 (Esp3I、CfrI、MboI)酶切并比较。结果 PCR扩增出 3株弓形虫的目的基因片段大小均在 5 0 0bp - 75 0bp之间 ,约 711bp。三种限制性内切酶酶切片段的大小均与理论值相符。 结论 弓形虫不同地理株GRA7基因具有高度保守性。
The genes encoding dense granule protein from different geographical strains of Toxoplasma gondii(RH, ZS 2,GT strains) were compared in this study. The GRA7 gene was amplified by PCR from the genomic DNA of different geographical strains of T.gondii, and the target gene was digested by restriction endonucleases(Esp3Ⅰ, CfrⅠ, MboⅠ) respectively.The experimental results showed that target gene fragments with size of about 711 bp (500-750 bp) in 3 different geographical strains were amplified by PCR, and the size of digested fragments produced by 3 restriction endonucleases were not significantly different, approaching the theoretical values. It concludes that the GRA7 genes from different geographical strains of T.gondii show high degree of conservation.
