摘要
报告了在碳酸胍体系中研究 6种 5′ 单磷酸核苷酸的分离工作 ,考察了分离条件对分离结果的影响。在最佳条件下 ,TMP、AMP、CMP、GMP、IMP、UMP等 6种核苷酸的线性范围分别为 :5~ 3 5 0、2~ 3 0 0、1 0~ 40、5~ 3 5 0、1 0~ 40 0和 1 0~ 40 0mg/L ;其检出限分别为 :1、0 .5、2、1、2和 2mg/L。初步探讨了胍基与不同核苷酸之间的相互作用及这种作用对毛细管电泳分离的影响。并将实验结果与普通碳酸盐体系作了比较。
s Six 5′-monophosphate nucleotides were separated by using guanidine carbonate as running electrolyte. The effect of separation conditions was studied. The Linear range, regression equation and detection limit were given. The linear range of six nucleotides was (mg/L): thymidine-5′-monophosphate (TMP): 5~350; adenosine-5′-monophosphate (AMP): 2~300; cytidine-5′-monophosphate (CMP): 10~400; quanosine-5′-monophosphate disodium salts (GMP): 5~350; inosine-5′-monophosphate disodium salt hydrate (IMP): 10~400; uridine-5′-monophosphate (UMP): 10~400. The detection limit was (mg/L): TMP: 1, AMP: 0.5, CMP: 2, GMP: 1, IMP: 2; UMP: 2. A comparision was made between the guanidine carbonate and Na 2CO 3-NaHCO 3 as running electrolyte.
