摘要
研究了从甜菜中分离高分子量核DNA的方法 .利用离心分离细胞核 ,经低融点琼脂糖块包埋、蛋白酶K原位裂解 ,结合低融点琼脂糖脉冲电泳去除淀粉制备高分子量核DNA .结果表明利用幼叶制备高分子量核DNA分子量高 ,更容易去除其中的淀粉 ;并且利用该方法得到的高分子量核DNA纯度高 ,适于部分酶切 .
A method of isolating HMW DNA from Beta vulgaris ssp. saccharifera nuclei was studied. It included isolation of purified nuclei by centrifugation, embedment of the nuclei in low-melting point agarose plugs, digestion in proteinase K in situ to release high molecular weight DNA, and isolation of purified HMW DNA from abundant starch using low melting agarose pulsed-field gel electrophoresis. The results indicated that it was easier to isolate HMW DNA from starch and acquired more HMW DNA using young green leaves than old ones. The quality of HMW DNA from Beta vulgaris ssp. saccharifera nuclei prepared by this method was greatly improved compared to traditional methods. The prepared DNA by this method was ready for partial digestion with restriction enzymes.
