四环素-精氨酸-甘氨酸-天门冬氨酸-酪氨酸的体内分布及其对骨代谢的作用 被引量:2

摘要 目的 探讨四环素-精氨酸-甘氨酸-天门冬氨酸-酪氨酸(RGDY)在小鼠体内的分布和在大鼠体内对骨代谢的影响。方法 同位素示踪方法检测四环素-RGDY在小鼠体内分布,骨组织计量学方法检测其对骨代谢的影响。结果 同位素示踪物在维甲酸造成的骨质疏松小鼠模型骨组织中的含量显著高于正常对照组。四环素-RGDY可以抑制去卵巢后骨质疏松所致骨小梁的丢失,表现在骨小梁体积和全部骨组织体积之比(TBV/TTV)、骨小梁体积和海绵骨体积之比(TBV/SBV)和平均骨小梁板厚度(MTPT)均较去卵巢组显著增加,平均骨小梁板间隙(MTPS)和骨小梁表面和体积之比(TBS/TBV)较去卵巢组显著减少(P<0.05)。与假手术组比较,四环素-RGDY治疗组TBV/SBV和平均骨小梁板密度显著减少,但TBS/TBV较之减少,MTPT较之显著增加(P<0.05);TBV/TTV和MTPS与之比较差异无显著。结论 四环素-RGDY能在骨组织聚集,其聚集能力不仅与四环素载体作用有关,而且与骨组织所处的代谢状态密切相关,四环素-RGDY可以抑制去卵巢后骨小梁的丢失。 Objective To investigate the distribution of tetracycline-arginine-glycine-aspartate-tyrosine (T-RGDY) in mice and its effect on bone. Methods 125I-labeled T-RGDY was studied for its distribution in mice and for its effects on bone by histomorphometry in ovariectomized rats. Results The 125I-labeled T-RGDY was more concentrated in the osteoporotic bone than in the normal bone. Compared with ovariectomy group, the morphologic index such as trabecular bone volume/total tissue volume (TBV/TTV), TBV/sponge bone volume (SBV), and mean trabecular plate thickness (MTPT) in T-RGDY group significantly increased (P < 0.05). As compared with sham operation group, MTPT significantly increased in T-RGDY group (P < 0.05), while TBV/SBV and mean trabecular plate density significantly decreased (P < 0.05), and TBV/TTV and mean trabecular plate spacing were almost the same as those in sham operation group (P > 0.05). Conclusion T-RGDY may concentrate in bone tissue to a certain degree, which is closely related with the status of bone remodeling. T-RGDY may inhibit the bone loss caused by ovariectomy.
