摘要
目的 :研究兔脑缺血再灌注损伤 (I/R)过程中海马组织磷脂酶A2 (PLA2 )活性的动态变化及川芎嗪的脑保护作用。方法 :采用双侧颈总动脉夹闭法 :先夹闭 2 0min造成脑缺血 ,松夹即为再灌注 ,建立兔脑缺血再灌注损伤模型。将兔随机分为 :缺血再灌注组 (I/R组 )、川芎嗪治疗组 (川芎嗪组 )和假手术对照组 (SOC组 )。应用pH酸度计检测海马组织PLA2 的活性 ;采用干湿重法、TTC染色法测定皮层脑组织含水量、梗死面积 ;光镜下观察脑组织病理变化。结果 :I/R组再灌注 1h、6h和 1 2h后海马组织PLA2 活性较SOC明显增高 (P <0 .0 1 ) ;川芎嗪组 ( 1h、6h、1 2h)PLA2 活性明显降低 ,与SOC组和I/R各相应时间点比较具有显著性差异 (P <0 .0 5 ,P <0 .0 1 ) ,且随再灌时间延长 ,PLA2 活性逐渐向正常水平恢复。同时川芎嗪组能显著降低脑梗死面积和脑含水量 ,脑组织病理变化也轻于I/R组。结论 :川芎嗪可通过降低脑组织PLA2 活性 ,改善脑循环 ,减轻脑缺血再灌注损伤 。
Objective: To observe the dynamic changes of phospholipase A 2(PLA 2) activity in the hippocampus structure in rabbits suffering from ischemia-reperfusion(I/R)damage,and to investigate the protective effects of ligustrazine. Methods:The cerebral lschemia model of rabbit was got by clipping bialatral carotide arteries for 20 minitues,and reperfusion was performed when the clips were released。Rabbits were randomly divided into sham operated control group(SOC),ischemia-reperfusion group(I/R)and ligustrazine group. PLA 2 activity in hippocampal structure were measured by using PH instrument. Dry-wet-weight way and TTC dye way were taken to measure content of water and clog area in cerebral tissure。The pathologic changes of brain tissues were observed by light-microscope。Rusults:After 1 hour,6 hour and 12 hour,PLA 2 activity had a more marked rise in I/R group than SOC group(P<0.01). PLA 2 activity did distinct drop in ligustrazine group (1h、6h、12h),compared with SOC and I/R group(P<0.05,P<0.01),PLA 2 activity came back to the control level following reperfusion time. The clog area and content water in brain tissue in ligustrazine group had a more marked drop and pathologic changes was lighter than I/R group。Conclusion: Ligustrazine can reduce PLA 2 activity,improve cerebral blood circulation and decrease brain damage in the I/R injury and play a protective effects。
