摘要
目的:内流性钾通道(Kir)2.1蛋白在大鼠脊髓和小脑的分布。方法:免疫荧光组织化学和免疫荧光双标记。结果:在脊髓灰质前角的运动神经元和白质的星形胶质细胞呈现 Kir2.1免疫反应活性强阳性。少数少突胶质细胞呈现Kir2.1和GS共存。在小脑蒲肯野氏细胞及纤维呈Kir2.1免疫活性强阳性。双标记显示近蒲肯野氏细胞旁的少突胶质细胞的Kir2.1蛋白和GS蛋白共存。结论:脊髓和小脑灰质和白质中的Kir2.1表达不一,这种分布可能与保持细胞外的K^+平衡、促进K^+的内流维持细胞兴奋性有重要关系。
Objective : To examine the distribution of the inwardly rectifying K+ channels Kir2.1 in the rat spinal cord and cerebellum. Methods : The immunofluorescence and double labeling irnmunofluorescence were performed on 10 um frozen sections of spinal cord and cerebellum fixed with 4% paraformaldehyde. Results : Kir2.1 immunostaining was revealed in neurons in the anterior horn and astrocytes in the spinal cord. To apply anti-glutamine synthetase antibody which is the mark of oligodendrocytes, double labeling anti-GS and anti-Kir2.1 showed 2 different proteins in few oligodendro-cytes. Meanwhile, Kir2.1 positive cells were viewed in Purkinje cells and fibers. Double labeling of anti-Kir2.1 and anti-GS showed 2 different proteins in perineuronal oligodendrocytes. Conclusion : These results reveal.that Kir2.1 is localized in different cells of spinal cord and cerebellum, suggesting that Kir2.1 may be involved in the transport of K+ in central nerve system, and playing a role in subserving inwardly K+ and controlling the excitability.
