摘要
目的 :观察镉对小鼠脾 T淋巴细胞增殖功能、c AMP(环磷酸腺苷 )、 IL- 2 (白细胞介素 - 2 )等的影响 ,探讨它们在镉的免疫毒性中作用和关系。方法 :对 BAL B/ C小鼠体外、体内染镉后 ,测定脾 T淋巴细胞增殖功能、IL- 2活性、c AMP含量等。结果 :体内、体外染镉均引起了小鼠脾 T淋巴细胞增殖功能、IL- 2水平有剂量 -效应关系的降低 ,两者的变化呈正直线相关。体内染镉引起 IL- 2水平对数的降低与 c AMP水平的升高呈负直线相关。体外染镉引起 T淋巴细胞增殖功能、IL- 2水平的降低与 c AMP水平升高呈负直线相关。结论 :镉有抑制小鼠脾 T淋巴细胞 IL- 2活性的免疫毒性作用 ;IL- 2在镉抑制 T淋巴细胞增殖活性中有重要作用 ;c AMP信使系统可能参与介导了镉对脾 T淋巴细胞增殖功能、 IL-
Objective:To study the relationship between effects of cadmium on proliferation and IL-2 activity and cAMP in mice lymphocytes.Methods:BALB/C mice were treated with CdCl_2 in physiological saline in vivo and in vitro,and then cAMP,IL-2 activity and proliferation or mice T lymphocytes were tested.Results:Treated with CdCl_2 ,the proliferation and IL-2 activity of mice splenic T lymphocytes were significantly dose-response lower than the control group.Logarithm IL-2 level and proliferation of mice splenic T lymphocytes were all suppressed simultaneously with positive line correlation between the both in vivo and in vitro.Treated with CdCl2 in vivo,cAMP level of mice splenic T lymphocytes were significantly dose-respose higher than the control group but there was no significant difference in vitro.Correlation analysis showed that in vivo logarithm of IL-2 level were negative line correlated with cAMP level of mice splenic T lymphocytes.Conclusion:It suggested that both in vivo and in vitro,cadmium executed immunotoxicological effects of suppressing IL-2 activity of mice splenic T lymphocytes,IL-2acitvity plays an important role in effects of cadmium on proliferation of mice splenic T lymphocytes.cAMP possibly takes part in mediating suppression of cadmium on IL-2 activity and proliferation of mice splenic T lymphocytes.
