樟子松成熟胚的离体培养与不定芽的形成 被引量:14

摘要 沙藏45d的樟子松种子,剥胚培养在5个系列17种培养基上.在6BA0.2~2.0(mg/L,单位下同)+NAA0.05~0.5以及6BA1.0+IAA0.2~0.5的激素配比范围内,可诱导胚形成不定芽.KT与NAA或IAA配合时无不定芽形成.不定芽分化部位明显受激素种类控制,6BA与NAA配合可诱导胚轴分化出不定芽,而6BA与IAA配合,则可从子叶顶端和基部诱导出不定芽。 This paper reports the regeneration of adventitious buds from mature embryos of P. sylvestris var mongolica. Embryos, excised ascpilically from seeds which were treated by stratification for 45 days, were cultured on 17 kinds of media. The adventitious buds arose from cxptant when growth regulators of 6BA 0.2~2.0mg/L +NAA 0.05~0.5mg/ L or 6BA 1.0mg/ L +IAA 0.2-0.5 mg/ L were supplemented to media. Noncbuds differentiated from embryos when KT was substituted for 6BA. The locations and patterns of adventitious buds differentiated from embryos were affected significantly by the kinds of growth regulators. When Basal media supplemented with 6BA and NAA, adventitious buds arose from hypocolyls. If TAA substituted for NAA, buds differentiated from the tips and bases of cotylcndons.
