摘要 目的对黄芪皂苷脂质体进行质量评价,测定黄芪皂苷脂质体的包封率。方法采用大孔吸附树脂分离脂质体与游离药物,用高效液相色谱-蒸发光散射检测器检测药物含量,计算包封率。结果该方法在0.026-1.229 mg·mL-1间线性良好(r=0.999 7),精密度小于1.24%,加样回收率在99.97%-101.67%之间,预饱和大孔吸附树脂柱对空白脂质体的平均回收率为100.16%,且能很好保留溶液中及脂质体制剂中游离药物。结论该方法具有操作简单,重现性好的优点。 OBJECTIVE: To evaluate the quality of astragaloside liposome and determine its entrapment efficiency. METHODS: Macroreticular resin was used to separate the free astragaloside from the liposomes and HPLC-ELSD was used to determine the entrapment efficiency. RESULTS: This method had good linearity in the range of 0.026-1.229 mg&middotmL-1 (r = 0.999 7). The precision RSD was less than 1.24%, the recovery of astragaloside with blank liposome was between 99.97%-101.67%, and the average recovery of blank liposomes through presaturated macroreticular resin was 100.16% with an excellent adsorption of free astragaloside. CONCLUSIONS: This method was simple and reproducible.
