摘要 目的 :研究黄芩茎叶总黄酮 (SSTF)对四氯化碳诱导的实验性大鼠肝纤维化的影响 ,并探讨其作用机制。方法 :6 0只大鼠随机分为 5组 ,正常组、模型组、SSTF(17 5mg/kg、35mg/kg、70mg/kg) 3个剂量保护组 ,采用四氯化碳 (CCL4)皮下注射 8周制备大鼠慢性肝纤维化模型 ,用SSTF灌胃给药 ,每天 1次 ,连续 8周 ,8周末处死全部大鼠。常规生化方法检测肝功能和肝组织Hyp含量 ,免疫组化检测肝组织转化生长因子 - β1(TGF - β1)和α -平滑肌肌动蛋白 (α-SMA)表达。H E染色和Van -Gieson染色观察肝组织病理形态学改变。结果 :SSTF保护组肝纤维化程度显著低于模型组 ,血清ALT活性和肝组织HYP含量明显减少 (P <0 0 5 ) ;CCL4诱导肝纤维化肝组织中TGF - β1、α-SMA表达增加 ,给予SSTF保护后可明显抑制肝组织中TGF - β1、α -SMA蛋白的表达。结论 :SSTF能有效减轻大鼠肝脏损伤和肝纤维化的形成 ,其机理可能是通过抑制TGF - β1在肝内的表达和星形细胞的活化 ,从而抑制了细胞外基质在肝脏的分泌和沉积 ,达到抗肝纤维化作用。 Objective :To study the effects of Scutellaria baicalensis stem-leaf total flavonoid(SSTF)on rat experimental liver fibrosis induced by CCL_4. Methods :All rats were divided into five groups:normal control,model group,SSTF there groups.Rats liver fibrosis model was induced by subcutaneous injection of CCL_4 twice a week for 8 weeks.During the course,rats were protected with SSTF(17.5,35,70 mg/kg)by stomach every day for 8 weeks all rats were sacrificed.Pahtological change of liver tissue observed under light microscope by H.E and Van-Gieson staining.The activity of serum alanine transaminase(ALT)and content of rat liver tissue hydroxyproline(Hyp)were measured.The expression of transforming growth facter β_1(TGF-β_1)and α-smooth muscle actin(α-SMA)in liver tissue was detected by immunohistochemistry. Result :SSTF could improve the pathological changes of liver tissue,the level of serum ALT and the contents of liver Hyp were significantly decreased.The expression of TGF-β 1 and α-SMA in the protective group were much lower than those in the CCL_4 group. Conclusion :SSTF has protective effect on liver injury and can inhibit liver fibrosis in rats induced by CCL 4,probably because it can depress the expression of TGF β 1 and the activation of hepatic stellate cell,then production of extracellular matrix were inhibited.
