摘要
目的 探讨复苏犬心肌细胞线粒体Ca2 + ATP酶活性及Ca2 + 含量的变化及其在复苏后心肌损伤中的作用。方法 电击致犬室颤 ,15分钟后进行标准复苏 ,测量心脏停跳 15分钟 ,复苏后 0 5小时、 2小时、 4小时心肌线粒体Ca2 + ATP酶活性及Ca2 + 离子含量。结果 与正常组相比 ,Ca2 + ATP酶活性于复苏后半小时显著下降 (P<0 0 1) ,复苏后 4小时恢复正常 (P >0 0 5 )。线粒体Ca2 + 含量与正常组相比 :复苏后半小时明显升高 (P <0 0 1) ,2小时达最高 ,复苏后 4小时仍未恢复 (P <0 0 1)。各实验组分别与相应的阴性对照组相比 ,结果与上述类似。结论 复苏犬心肌线粒体Ca2 + ATP酶活性及Ca2 +
Objective To investigate changes of mitochondria Ca^(2+) ATPase activity and Ca^(2+) content in myocardium before and their effects on myocardial injury after resuscitation in post-resuscitation dogs.Methods Dogs were given electrical shock with ventricular fibrillation/cardiac arrest (CA) and normally resuscitated after 15 minutes. Ca^(2+) ATPase activity and Ca^(2+) content of myocardial mitochondrion were measured at 15 minutes after CA, 0.5h, 2h, 4h post-resuscitation respectively.Results Ca^(2+) ATPase activity of myocardial mitochondrion decreased significantly (P<0.01= compared with that of the normal group after 30 minutes of CA and restored normally after 4h post-resuscitation(P>0.05). Mitochondrial Ca^(2+) content increased significantly(P<0.01) after 0.5h and reach peak values after 2h post-resuscitation, not yet restored after 4h post-resuscitation(P<0.01). The result of each experimental group's index compared with responding sham control group's is similar to the result of each experimental group's compared with the normal group's.Conclusion Changes of myocardial mitochondrion Ca^(2+) ATPase activity and Ca^(2+) content could take participate in myocardial injury post-resuscitation.
