摘要 目的比较两种不同方法分离脐血造血干细胞的结果。方法采用Ficoll法、HES法分离脐血单细胞,比较两种方法优点。结果HES法所得MNC产率(79.31±2.68)%、GM-CFU数(60.92±9.49)均显高于Ficoll分离法所得MNC产率(41.72±4.19)%、GM-CFU数(31.17±5.26) (P<0.01)。该研究采用S法分离脐血造血干细胞对HLA相合的相关脐血移植已成功重建造血。结论HES分离法病原菌污染机,且单个核细胞、造血干/祖细胞的回收率较高,对重建造血无不良影响,是一种较好的分离方法。 Objective: To compare the results of umbilical cord blood hematopoietic stem cells separated by two different methods. Methods: Umbilical cord blood mononuclear cells were separated by Ficoll or Hydroxethy1 strach (HES), and then the two methods were compared. Results: The number of MNC and GM-CFU separated by HES were (79.31±2.68)% and (60.92±9.49) and the number of MNC and GM-CFU separated by Ficoll were (41.72±4.19)% and (31.17±5.26) respectively. Statics analysis shows that both number of MNC and GM-CFU separated by HES is significant different than that by Ficoll (P<0.01). Fruthemore, the hematopoietic stem cells separated by HES was successful used in a HLA matched cord blood stranplantation. Conclusions: Cord blood separated by HES has a low frequency of pathogenic bacteria contamination and high recovery of mononuclear cell and stem progenitor cells. It has no negative effect on repopulating. So it is a preferable measure of separation.
