摘要
目的研究日本血吸虫极低密度脂结合蛋白重组抗原(reSVLBP)在血吸虫病诊断中的应用价值及现场推广前景。方法以reSVLBP为抗原建立reSVLBP-ELISA,检测血吸虫病人及其它寄生虫感染者血清中特异性抗体,并与常规SEA-ELISA进行平行检测和比较。结果reSVLBP抗原和SEA对血吸虫的敏感性分别为90.5%和97.3%(P>0.05),两者对血吸虫的特异性分别为100%和77.0%(P<0.05);reSVLBP与肺吸虫、姜片虫血清未见交叉反应,与华支睾吸虫、蛔虫、鞭虫感染者血清的交叉反应率分别为20.0%、20.0%、10.0%。SEA对肺吸虫和姜片虫病人血清的交叉反应率分别为29.4%和16.7%,与华支睾吸虫、蛔虫、鞭虫感染者血清的交叉反应率分别为26.7%、30.0%、20.0%。结论reSVLBP作为诊断抗原具有很高的特异性,与其它寄生虫的交叉反应少,敏感性与SEA相近,在血吸虫病诊断中具有一定的应用前景。
Objective To explore the potential and the prospect of recombinant specific very low density lipoprotein binding protein (reSVLBP) in immunodiagnosis of schistosomiasis in the field. Methods A reSVLBP ELISA was developed to detect the specific antibodies in serum samples from schistosomasis patients using reSVLBP as antigen, paralleling with SEA ELISA which used soluble egg antigen(SEA) to detect antibodies against SEA. Results The sensitivities of reSVLBP and SEA were 90.5%and 97.3%(P >0.05) respectively, and the specificities were 100%and 77.0%(P< 0.05) respectively. It showed that no cross reaction between reSVLBP and sera from Paragonimus and Fasciolopsis. The rates of cross reaction between reSVLBP and sera from Clonorchiasis, Ascariasis and Trichuriasis were 20%, 20%and10%respectively, while the cross reaction rates between SEA and sera from Paragonimus, Fasciolopsis, Clonorchiasis, Ascariasis and Trichuriasis were 29.4%, 16.7%, 26.70%, 30%and 20%respectively. Conclusion Although the sensitiveity of the reSVLBP ELISA is slightly lower than SEA ELISA, it is more specific and less cross reaction with sera from patients infected other parasites. It suggested that the reSVLBP had potential application prospect in schistosomasis diagnosis.
