摘要
目的 建立人胃癌细胞株SGC790 1和MKN4 5裸鼠腹腔内转移模型。方法 体外培养低分化胃癌细胞SGC790 1和MKN4 5 ,分别接种于裸鼠右后肢皮下 ,形成移植瘤后分别制备SGC790 1和MKN4 5的肿瘤悬浮液 ,分别将 0 .2mL 1× 10 6、1× 10 7和 1× 10 8浓度的肿瘤悬浮液直接接种于裸鼠腹腔内或行腹部小切口注入裸鼠腹腔。分别于接种后 2、3和 4周开腹 ,了解不同浓度、不同接种方法和不同开腹时间裸鼠腹腔内肿瘤种植的情况。结果 以 (1× 10 7) / 0 .2mL浓度的SGC790 1和MKN4 5的肿瘤悬浮液经裸鼠上腹部小切口直接腹腔内接种后 4周 ,可在裸鼠腹腔内形成直径约为 (3.4± 0 .83)mm(n =10 0 )且大小较为均一的腹腔内转移结节。结论(1× 10 7) / 0 .
Purpose: To develop the animal model with human gastric cancer metastasize to intraperitoneal cavity. Methods: Two human gastric cancer cell lines SGC7901 and MKN45 were cultured in vitro and transplanted in the right hind legs of nude mice percutaneously. The tumor suspension both SGC7901 and MKN45 were made to inject to the nude mice peritoneal cavity by direct puncture or making a small incision in different concentration[(1 × 106)/0.2 mLinput commas (1 × 107)/0.2 mL and (1 × 108)/0.2 mL]. After 2 weeks, 3 weeks and 4 weeks respectively, the peritoneal cavity was reopened to investigate the peritoneal transplantation condition in different tumor suspention, different methods and different timing. Results: 4 weeks after injection into peritoneal cavity through a small incision in nude mice by tumor suspension of SGC7901 as well as MKN45 in (1 × 107)/0.2 mL, about (3.4 ± 0.83)mm(n = 100) diameter uniform metastasis node can be found in peritoneal cavity. Conclusions: The gastric cancer peritoneal metastasis nude mice model can be achieved by injection of human gastric cancer cell line into nude mice peritoneal cavity in certain concentration suspention.
