摘要 目的 建立人类雌激素受体 (estrogenreceptor,ER)和孕激素受体 (progesteronereceptor,PR)Westernblot检测方法 ,并对不同乳腺癌细胞株ER、PR表达情况进行测定。方法 取不同乳腺癌细胞株对数生长期细胞提取总蛋白 ,分别用ERα单克隆抗体 1D5和PR单克隆抗体PgR6 36以Westernblot方法对ER、PR的表达情况进行检测 ,并与同期固定细胞的流式细胞术 (FCM)检测结果进行比较。结果 T 4 7d、MCF 7、ZR 75 1细胞ERα的Westernblot检测可得到清晰且分子量正确的条带 ,T 4 7d、ZR 75 1细胞可见清晰且分子量正确的PR条带 ,同期固定细胞ER、PR阳性表达的FCM检测结果与Westernblot检测结果一致。结论 Westernblot检测ER、PR的方法是可行的。 Objective To establish Western blot analysis of human estrogen receptor (ER) and progesterone receptor (PR), and detect the expression of ER and PR in different human breast cancer cell lines. Methods After extracting total proteins from different exponentially growing cell lines, the expression of ER and PR was detected by Western blot with 1D5 and PgR636, monoclonal antibody to ERα and PR, respectively. And the results were compared with those of fixed cells detected by flow cytometry. Results Clear and correct ERα bands were observed by Western blot analysis in the lysates from cell lines T-47d, MCF-7 and ZR-75-1. In addition, clear and correct PR bands were observed in the cell lines T-47d and ZR-75-1. Positive expression of ER and PR in different fixed cells detected by flow cytometry was the same with that analyzed by Western blot. Conclusion Western blot analysis of ER and PR are feasible.
