整合素α_5亚基及纤维粘连蛋白与肺腺癌细胞凋亡关系的实验研究

摘要 目的 探讨整合素α5亚基及纤维粘连蛋白与人肺腺癌A5 4 9细胞凋亡之间的关系。方法 无血清培养 ,以抗整合素α5亚基单克隆抗体 (SAM - 1)封闭A5 4 9细胞表面的整合素α5亚基 ,阻断其与纤维粘连蛋白的粘附作用。结果 SAM - 1封闭整合素α5亚基后细胞凋亡明显增强。各实验组AI值与对照组相比差异有显著性意义(P <0 0 1) ,在一定范围内 ,呈剂量、时间依赖关系。结论 整合素α5亚基与纤维粘连蛋白可能抑制体外无血清培养诱导的A5 4 Objective To study the relationship among integrin α 5 subunit,fibronectin and the apoptosis of lung carcinoma cells in vitro.Methods Human lung adenocarcinoma A549 cells was cultured on the petri dishes coated by fibronectin in serum-free medium in vitro, then was treated with the specific monoclonal antibody to integrin α 5 subunit (SAM-1) at different concentrations to disrupt cell-fibronectin interaction.Results Blocking the function of integrin α 5 subunit can distinctively induce apoptosis of A549 than the control group (P<0.01).In some range, the cell apoptosis index (AI) increased with the dose of antibody and culture time (P<0.01).Conclusions Integrin α 5 subunit and fibronectin can delay the apoptosis triggered by serum deprivation in vitro. It proposed that Integrin α 5 subunit,fibronectin and the of human lung adenocarcinoma A549 cells were correlative.
