摘要
[目的 ]建立肿瘤化疗免疫抑制动物模型 ,为建立保健食品减轻化疗毒副作用功能的评价方法提供科学依据。 [方法 ]给予C57BL/6J正常小鼠和荷瘤小鼠腹腔内注射不同剂量的环磷酰胺 (CP) ,隔日连续腹腔注射 4~ 5次 ,观察对小鼠免疫学指标及肝肾功能的影响。 [结果 ]正常小鼠和荷瘤小鼠腹腔注射 2 0、5 0、10 0mg/kg体重的CP后 ,各剂量组均出现不同程度的免疫抑制和肝肾功能损伤。①正常小鼠 2 0mg/kg体重以上剂量组 ,白细胞计数、抗体生成细胞以及巨噬细胞功能显著下降 ,谷草转氨酶 (AST)升高 ;5 0mg/kg体重剂量组的脾脏指数下降 ;5 0mg/kg体重以上剂量组天然杀伤细胞(NK)细胞活性均出现显著下降 ,以及谷丙转氨酶、尿素的升高。②荷瘤小鼠 2 0mg/kg体重以上剂量组白细胞计数、巨噬细胞功能显著下降 ,5 0mg/kg体重以上剂量组脾脏指数、NK细胞活性、抗体生成细胞功能均出现显著下降 ,以及谷草转氨酶、谷丙转氨酶、尿素的升高。③正常小鼠注射 60mg/kg体重的CP后 ,血清白细胞介素 2及肿瘤坏死因子均出现显著下降 ;荷瘤小鼠注射 60mg/kg体重的CP后 ,血清白细胞介素 2出现显著下降。 [结论 ]给予C57BL/6J正常小鼠和荷瘤小鼠腹腔注射 2 0 5 0mg/kg体重的CP ,隔日连续 4 5次 ,即可建立免疫抑制动物模型。建议采?
Objective To establish immunosuppressive animal model and to study the assessment method for function food alleviating side effect of chemotherapy.[Methods] Different doses of cyclophosphamide(CP) were given to normal and tumor-bearing C57BL/6J mice every other day during 10 days.Immunological indices,liver and kidney functions were examined.[Results] 1.In 20 mg/kg bw group of normal mice,the white blood cells,antibody-producing cells and macrophage function declined and the level of serum AST increased significantly. In 50 mg/kg bw and 100 mg/kg bw groups of normal mice,NK cell activity descended and the levels of AST,ALT and BUN ascended significantly. 2.In all tumor-bearing mice groups,the white blood cells and macrophage function declined significantly. In 50 mg/kg bw and 100 mg/kg bw groups of tumor-bearing mice,spleen index,NK cell activity and antibody-producing cell function descended and the levels of AST,ALT and BUN ascended significantly. 3.After injection of 60mg/kg bw of CP,the levels of IL-2 and TNF-α in normal mice and the level of IL-2 in tumor-bearing mice decreased significantly. [Conclusion] Immunosuppressive animal model has been established by intraperitoneal injection of CP at the doses of 20-50 mg/kg bw to both normal mice and tumor-bearing mice.
