摘要
目的 :观察异氟醚麻醉后大鼠中枢神经系统c-fos基因的表达 ,了解异氟醚在中枢神经系统的作用部位 ,探讨吸入全麻机理 .方法 :SD大鼠 4 0只 ,雌雄不拘 ,体重 (178 6± 18 0 )g ,日龄 (80± 15 )d ,随机分为对照组、 0 6 %异氟醚组 ,1 2 %异氟醚组和 1 8%异氟醚组 ,每组 10只 .实验组分别吸入 0 6 % ,1 2 %和 1 8%的异氟醚 1h ,对照组除不吸入异氟醚外 ,其余条件完全同实验组 .停止吸入后 ,每只动物腹腔注射 3%戊巴比妥钠 (4 0mg/kg)麻醉后开胸 ,经升主动脉灌注生理盐水 10 0mL洗去全身血液 ,继之灌注多聚甲醛液 (0 1mmol/L ,pH 7 4 ,含 4 %的多聚甲醛 ) 2 5 0mL ,固定 90min .取全脑石蜡包埋 ,切片 (片厚 5 μm) ,应用c-fos基因免疫组织化学方法 ,低倍显微镜 (× 10 )下观察染色阳性细胞的分布情况 ,高倍镜 (× 2 0 )下观察鉴别Fos染色免疫阳性细胞并作阳性细胞记数 .结果 :对照组中未发现Fos阳性细胞 ,实验组大鼠的皮质、丘脑、中脑、桥脑、延髓、小脑可观察到Fos蛋白阳性神经元 ,与对照组相比P <0 0 5和P <0 0 1,这些神经元的分布局限于某些区域 ;同时发现Fos蛋白阳性神经元随吸入异氟醚浓度的增加而增多 ,实验组间相比P <0 0 5和P <0 0 1.结论 :异氟醚能诱导大鼠皮质、丘脑、中脑、桥脑、延髓?
Objectives: To investigate the expression of c-fos oncogene in rats, central nervous system following Isoflurane anesthesia, sdudy Isoflurane action sites in CNS, and discuss general anesthetic mechanisms of inhalation anesthetics. Methods: 40 adult SD rats, weight (178.6±18.0)g, were randomly divided into 4?groups: control group (n=10), 0.6% Isoflurane group (n=10),1.2% Isoflurane group (n=10), 1.8% Isoflurane group (n=10). Experimental groups inhalated 0.6%,1.2%,1.8% Isoflurane for 1?h. The control group underwent the same steps except without inhalating Isoflurane. After 1?h, each rat was anesthetized by intraperitoneal injection pentobarbital (40?mg/kg), then opened, chest and perfused normal saline 100?mL as well 4% poly-formalindehyde 250?mL. 90?min later the whole brain was paraffined-inbeded and sliced. C-fos oncogene immunohistochemical technique was employed to observe and count on the Fos-like immunoreactive (FLI) cells. Results: In control group, there was no FLI cell seen.The experimental groups could see FLI cells in the cortex, thalamus, midbrain, bridgebrain, medulla oblongata and cerebellum. Compared to control group P<0.05 and P<0.01, those FLI cells were limited in some region. Meanwhile finding FLI cells were increased with Isoflurane concentration raised, and among experiment groups the difference P<0.05 and P<0.01. Conclusions: (1) Isoflurane can induce expression of c-fos oncogene in the cortex, thalamus, midbrain, bridgebrain, medulla oblongata and cerebellum of rats .(2) The cortex, thalamus, midbrain, bridgebrain, medulla oblongata, and cerebellum are the action sites of Isoflurane.
