摘要
目的观察雷公藤甲素对胶原诱导的关节炎(CIA)模型大鼠膝关节腔液肿瘤坏死因子(TNF)-琢、白细胞介素(IL)-10含量和对滑膜组织核转录因子资B(NF-资B)表达与活性的影响,探讨雷公藤治疗类风湿关节炎(RA)的可能作用机制。方法于Wistar大鼠背部及尾根部多点皮内注射牛Ⅱ型胶原乳剂制作CIA动物模型,经过雷公藤甲素治疗后,通过酶联免疫吸附(ELISA)法检测大鼠膝关节腔液TNF-琢和IL-10含量,免疫组织化学染色(SP)检测NF-资B的表达和凝胶电泳迁移分析法(electrophoreticmobilityshiftassay,EMSA)测定NF-资B活性。结果与正常对照组大鼠相比,CIA模型大鼠关节腔液TNF-琢含量升高(P<0.05),IL-10含量降低(P<0.05),膝关节滑膜组织NF-资B的表达和活性增加(P<O.05),经过雷公藤甲素治疗后以上指标恢复至正常水平(P>0.05)。结论雷公藤甲素可能通过下调滑膜细胞NF-资B的表达与活性,降低炎性因子的表达,升高抑炎因子的表达而治疗RA。
Objective To explore the possible role of triptolide in rheumatoid arthritis (RA) by detecting the expression and activity of nuclear factor kB (NF-kB) in the synovium of collagen-induced arthritis (CIA) rats. Methods The experimental Wistar rat model of CIA was set up by intradermal injection of bovine type Ⅱ collagen emulsion. The success rate of models set-up was evaluated by arthritis index (AI). Rats were grouped randomly into three groups: normal, control and triptolide group. The expressions of TNF-α and IL-10 in synovial fluid were detected by ELISA, the expression of NF-kB in synovium was detected by immunohistochemistry method and the activity of NF-kB in synovium was detected by electrophoretic mobility shift assay (EMSA). Results Compared with normal group, the expression of TNF-α in synovia was increased (P<0.05) and IL-10 was decreased (P<0.05), the expression and activity of NF-kB (P<0.05) in synovium was increased in CIA control group. There was no statistical difference in above indexes between normal group and triptolide group (P>0.05). Conclusion Triptolide may play a protective role in RA by downregulating the expression and activity of NF-kB in synovium.
